i would like to ask any brother or sister that  is there any rule in islam that stops a girl from marriying if she has an unmarried older sister?

There is no such thing in Islam. That's just culture.

thats what i thought but people find excuses not to  get married to the person. i relly don't like when they mix culture with religion

Salam,

In a certain degrees, I agree. However we shouldn't anti culture. Culture is something that make the live and other things become beautiful. However religion should correct culture that against the core value of the religion. So religion and culture should and will interact and affect each other in "positive" way.
 
During the Prophet Muhammad's times, slavery and alcoholism were everywhere, the teaching of Islam is not in-line with slavery and alcoholism, so gradually slavery and alcoholism were disappear from the muslim community.
 
An example of culture, that do not against religion is muslim's dresses.
 
Muslim's dresses from many different parts of the world have different styles and colors. For example middle eastern men like to wear white kufi (hat) but south east asian like to wear black kufi. Middle eastern women like to wear black hijab, the african like brown, south east asian like colorful or white hijab. Indo-Pakistani men like to wear salwar kameez, Indonesian-Malaysian like to wear sarong.  And now, muslim men everywhere, they wear western styles of dresses (pants, shirts and blazers for formal and jeans and tshirt for casual). That's OK, these are cultures that do not against any of Islamic value.
